Isolation and Characterization of Native Strains of Leuconostoc oenos from Washington State Wines

Native strains of Leuconostoc oenos, isolated from commercial Washington state wines, were characterized and compared to commercially available strains with regards to growth at different pH, SO2, and temperatures. Strains were also inoculated into a Washington State wine (Merlot) to evaluate their ability to induce MLF. Commercial wine samples were streaked on solidified modified apple Rogosa and Gram (+) cocci colonies in pairs and chains were subcultured and identified as strains of L. oenos. Most strains grew well at pH 3.3, 3.5, and 4.5, in 15 mg/L total SO2 (pH 3.5), and at 25°C and 30°C. None of the strains were able to grow at pH 2.9. Growth was retarded at 30 mg/L SO2 with strains WS-18 and WS-19A inhibited the greatest. Several native strains including WS-6A and WS-22 induced faster MLF in the Merlot wine compared to the commercial strains ML-34 and MCW.
