Effect of Benzyladenine Concentration and Double-Phase Culture System on In Vitro Multiplication of Adult Albariño Plants

The effect of varied culture conditions were studied for the in vitro establishment and multiplication of nodal segments from adult field-grown plants of Vitis vinifera L. cv. Albariño (clone Maceira). Benzyladenine (BA) concentration in the culture medium and a double-phase culture system for shoot multiplication and elongation both had significant effects on success. Optimal multiplication in solid medium was obtained using 22.2 µM BA, whereas elongation was greatest in the presence of 2.2 µM BA. The multiplication index, which reflects both axillary shoot multiplication and shoot elongation, was greatest with 8.8 µM BA. Shoot multiplication and elongation were improved when a double-phase (a layer of liquid medium over solid medium of the same composition) culture system was used, following a similar kinetics to that observed for solid medium. The highest cytokinin concentration for the multiplication index was 8.8 µM BA, representing more than a 2-fold increase over the index obtained with solid medium. Shoot multiplication and elongation were stable either in solid medium or in a double-phase culture system for at least five subcultures in the presence of all cytokinin concentrations tested, although further work is necessary to minimize the hyperhydricity observed in some of the cultures.
