Abstract

A modification of the enzyme-linked immunosorbent assay using the F(ab')2 antibody fragment [F(ab')2 ELISA] was developed for the detection of grapevine fanleaf (GFLV) and grapevine leafroll (GLRV-type III) viruses. The protocol was sensitive and rapid. Sensitivity of the test was further enhanced by pre-mixing the detecting antibody (probe) and the enzyme-conjugated protein A (anti-probe). Due to the acidic nature of grapevine tissues, different extraction buffer compositions were also evaluated.
